Title: First-in-class Claudin 18.2 targeting radiopeptide, 64Cu-PD-29875 and 225Ac-PD-
29875, exhibit promising theranostic potential in pancreatic and gastric cancer xenograft
models

About PD- 29875
PD-29875 is a macrocyclic peptide targeting Claudin 18.2 (CLDN18.2) discovered using
PeptiDream’s proprietary PDPS® technology and further optimized through in vivo imaging
and efficacy studies conducted at PDRadiopharma. CLDN18.2 is highly expressed in a variety
of solid tumors, including gastric cancer, esophageal cancer, pancreatic cancer and lung
adenocarcinoma. It has been reported to regarded as an attractive molecular target for both
cancer diagnosis and therapy. PD-29875 is being developed as a theranostic platform through
labeling with the diagnostic radionuclide 64Cu and the therapeutic radionuclide 225Ac. a Phase
0 clinical study of 64Cu-PD-29875 is currently underway in patients with gastric cancer and
GEJ cancer to evaluate its safety, pharmacokinetics, and tumor uptake. The program was
adopted by Japan Agency for Medical Research and Development (AMED) as part of the
“Practical Research for Innovative Cancer Control” and received funding support from AMED
in 2024.
